Research Interests

The long-term goal of our laboratory is to determine the biological role of Parotid Secretory Protein (PSP; BPIFA2) as an oral and systemic anti-inflammatory protein. PSP is expressed in salivary glands, oral epithelial cells and saliva and its expression is up-regulated by bacteria and TNF. An in silico structural analysis of PSP revealed that it may be similar to Bactericidal/Permeability Increasing protein (BPI) and Lipopolysaccharide (LPS)-Binding Protein (LBP). Indeed, recent evidence from our laboratory suggests that PSP binds LPS and prevents systemic inflammation in vivo. Guided by the location of active domains in BPI and LBP, several PSP peptides were designed and found to exhibit both antibacterial and anti-endotoxin activities in vitro and in vivo. Modified PSP peptides have been designed that show enhanced antibacterial activity and selectivity for certain bacterial species. Future goals include understanding the biological activity of PSP in vivo and optimization of PSP peptides as selective antimicrobial peptides with low host toxicity.
